Isomerase Launches Isochassis – Microbial Hosts for Scalable Protein Production

Isomerase has today announced the release of its Isochassis platform, a comprehensive collection of proprietary engineered microbial hosts designed to meet industrial recombinant protein and biologics production needs. Combined with in-house high cell density fermentation development services, yields can be maximised allowing for scalable production.

Our platform hosts include Escherichia coliPichia pastoris, as well as Bacillus subtilis, and will continue to grow as Isomerase invests in internal research and fermentation development services to remain at the forefront of enzyme production technology.

Why the Isochassis platform?

  1. Versatile and Customizable – Our team brings extensive experience collaborating with partners across various sectors, enabling us to effectively identify the optimal host for your project needs.
  2. Evaluation and Optimisation – Each partner can evaluate various combinations of system components enabling a precise understanding of productivity. Optimal systems can therefore be selected from the outset.
  3. Ownership – Strains made for your target belong to you with full commercialization rights. No downstream fees or royalties.
  4. Integrated Solutions – IsoChassis is an integral part of the IsoZym platform, which enables seamless protein development from sequence to scalable production as part of our fermentation development services, eliminating the need for complex and time-intensive technology transfers between providers.

Which microbial host suits my requirements?

Escherichia coli – Extensive selection derived from the widely utilized BL21 DE3.
  • BL21 DE3 lacY deletion mutants for tunable induced expression.
  • Protease-deficient strains.
  • Option for co-expressed chaperones, disulfide bond formation and rare codon expression.
  • Low temperature recombinant protein expression (e.g. 12 oC).
  • Improved toxic and low-yielding protein expression.
  • Metabolic and chromosomal engineering.
  • Periplasmic secretion and heterologous protein isolation using a range of secretion tags.
  • Variety of replicative high and low copy vectors.
  • Selection of constitutive and inducible promoters using various inducers.
Pichia Pastoris – Engineered for high level intracellular and extracellular protein production.
  • GRAS
  • Controlled expression in methanol-induced systems (Mut+ and MutS variants).
  • Reduced protein degradation with protease-deficient strains.
  • Wide range of secretion tags available.
  • Increased plasmid retention without the need for antibiotics with auxotrophic mutants.
  • Methanol-free expression system.
Bacillus subtillis – engineered strains derived from Bacillus subtilis Marburg.
  • GRAS
  • Suite of replicative and integrative vectors.
  • Protease-deficient strains and sporulation mutants.
  • Constitutive and inducible promoters.
  • Range of protein secretion tags.
